What it is used for: Hypertension.
Indications as listed by the manufacturer: Amlodipine Besylate is indicated for the first line treatment of hypertension and can be used as the sole agent to control blood pressure in the majority of patients. Amlodipine Besylate is indicated for the first line treatment of myocardial ischemia whether due to fixed obstruction (stable angina) and/ or vasospasm/vasoconstriction (prinzmetal’s or variant angina) of coronary vasculature.
How it works: The mechanism of the antihypertensive action of amlodipine is due to direct relaxant effect on vascular smooth muscle. The precise mechanism by which amlodipine relieves angina has not been fully determined but amlodipine reduces total ischemic burden by the following two actions. Amlodipine dilates peripheral arterioles thus, reduces the total peripheral resistance (after load) against which the heart works.
Active ingredient: Amlodipine Besylate.
Reported side effects: Amlodipine Besylate is well tolerated; the most commonly observed side effects were headache, edema, fatigue, nausea, flushing and dizziness.
Known drug interactions: Azole antifungals , erythromycin , cyclosporins , sildenafil , rifampicin , dantrolene , simvastatin , tacrolimus , other antihypertensives , grape fruit juice.
Adults : For both hypertension and angina, the usual initial dose is 5mg amlodipine once daily which may be increased to a maximum dose of 10mg depending on the individual patient’s response. Children : Under 6yr , not recommended. Hypertension : 6-12yr, initially 100-200mcg/kg once daily; if necessary increase at intervals of 1-2wk upto 400mcg/kg once daily; max. 10mg once daily; 12-18yr, initially 5mg once daily; if necessary increase at intervals of 1-2wk to max 10mg once daily. Always consult with your your doctor or pharmacist for dose adjustments.
