What it is used for: Antidote.

Indications as listed by the manufacturer: It is used in cancer treatment (cytotoxic therapy) as an antidote to reduce the toxicity (poisonousness) and effect of certain cancer drugs, the so-called folic acid antagonists (e.g. methotrexate).

How it works: Leucovorin is folic acid in its active (reduced) form, so it allows nucleic acid synthesis to proceed even in the presence of methotrexate. Leucovorin can also compete with methotrexate for the same transport processes into the cell .

Active ingredient: Leucovorin Calcium.

Reported side effects: Uncommon (may affect up to 1 in 100 people) : * Fever Rare (may affect up to 1 in 1,000 people) * Insomnia, restlessness and depression after high doses * Increase in the frequency of fits in epileptics * Stomach and intestinal complaints (gastrointestinal complaints) after high doses Very rare (may affect up to 1 in 10,000 people) : * Allergic (anaphylactic and anaphylactoid) reactions and hives (urticaria), ranging up to lifethreatening shock.

Known drug interactions: 5-fluorouracil , Methotrexate , folic acid antagonist (e.g. cotrimoxazole, pyrimethamine), phenobarbital, primidone, phenytoin and succinimide.
