How it works: Docetaxel is an antineoplastic agent that acts by disrupting the microtubular network in cells that is essential for mitotic and interphase cellular functions. Docetaxel binds to free tubulin and promotes the assembly of tubulin into stable microtubules while simultaneously inhibiting their disassembly. This leads to the production of microtubule bundles without normal function and to the stabilization of microtubules, which results in the inhibition of mitosis in cells. Docetaxel's binding to microtubules does not alter the number of protofilaments in the bound microtubules, a feature which differs from most spindle poisons currently in clinical use.
Active ingredient: Docetaxel.
Reported side effects: Irritation, pain, swelling or colouring around the needle during infusion • high temperature • Stomach pain or discomfort • feeling sick, upset stomach or vomiting • mild diarrhoea • constipation • inflammation of the food pipe (oesophagus) • whitening or darkening of the skin or nails • loosening of the nails • unusual hair loss or thinning • joint pain or swelling • frequent infections with fever, severe chills, sore throat or mouth ulcers - especially 5-7 days after receiving a docetaxel infusion • sore red mouth or vagina or swelling in these areas • severe diarrhoea • tiredness, headaches, being short of breath when exercising,dizziness and looking pale • coughing • change in the rhythm or rate of your heart beats (palpitations) • muscle pain or cramps • flushed, dry skin, irritability and confusion.
Known drug interactions: Ciclosporin , Erythromycin, clarithromycin and telithromycin , Ketoconazole, itraconazole , voriconazole , nifedipine , dexamethasone , phenobarbitone , ritonavir, indinavir, nelfinavir and saquinavir , nefazodone
Adults : 60-100mg/m2 IV over 1 hr every 3wk. Dosage adjustment may be required. Non-Small Cell Lung Cancer : 75mg/m2 1 hr after doxorubicin 50mg/m2 and cyclophosphamide 500mg/m2 every 3 wk x 6 courses. Dosage adjustment may be required. Children : Not Recommended. Always consult with your doctor or pharmacist for dose adjustments.
